Why Is Profiling for Aggregation Important when Selecting Your Lead Molecule?

Aggregation is a serious concern with large molecule therapeutics. Aggregated molecules are highly immunogenic, meaning that they cause aberrant activation of the immune system. This can result in drug reactions, side effects, and ultimately make a medication unsafe for administration.
 
Concentrating samples to a therapeutic dose can cause molecules to destabilize and clump or aggregate together. Conditions required for administration, such as buffer, temperature, transport, and storage can cause large molecules such as proteins to unfold and stick together or aggregate. Optimization of leads for manufacturing often requires a combination of modifications to both drug and buffer conditions to ensure stability across a wide range of conditions that would be encountered in the lifetime of a drug sample.
